Biomimetic pharmaceuticals are commonly used to treat prophylaxis of pathological conditions which are mediated by lymphocytes, NK cells, and immune cells. For instance, immunoglobulin products have been used since the early 1950’s to treat immune deficiency disorders. Intravenous immune globulin (IVIG) has been more recently developed for treatment of autoimmune diseases like idiopathic thrombocytopenic purpura. These treatment methods have had shortcomings though including common inadequate sterility, impurities, and infectious agents. Gliknik, Inc. is a biopharmaceutical company focused on creating new therapies for immune disorders.
Inspired by the previous success – although limited – with IVIG, Gliknik began development o f their own immunoglobulin based therapy. It involved the fusion of natural human protein fragments to create orderly and multimerized immunoglobulin Fc compositions. These can then be presented to immune cell receptors. Their method describes a way to achieve rational molecular design for human IVIG replacement and uses recombinant and biochemical creations of these biomimetics. The biomimetic hIVIG are more efficient at multimerization and therefore more efficient at binding to Fc gamma receptors. The replacement compounds have shown promise as a treatment option for autoimmune diseases, inflammatory diseases, cancer, and sepsis.
