Some studies show that tumor infiltrating lymphocytes (TIL) can successfully mediate durable regression of metastatic melanoma. However, the same success has not been seen in other tumors as they have microenvironments capable of suppressing the TIL activity. Genetically altering the lymphocytes to recognize molecular targets on the tumors have helped to achieve regression in hematologic tumors. Unfortunately, a marker and molecule combination has not been identified in all solid tumors. Intima Bioscience, Inc. has developed a procedure to target gastrointestinal tumors with genetically modified TIL.

Lymphocytes are white blood cells that are also one of the body’s main types of immune cells. TIL are capable of infiltrating a tumor, though their access may be restricted. The TIL are taken from a donor and subjected to genetic modification using a CRISPR system. The TIL have been engineered to disrupt a specific gene so that Cytokine Inducible SH2 Containing Protein (CISH) protein function is suppressed. CISH controls T cell receptor (TCR) signaling. This modification allows TIL to target and infiltrate the tumor. The TIL are now functional for the targeted demise of the tumor.
