Pennsylvania Patent of the Month – February 2021

Thyroid hormones (TH) play a critical role in growth and development as well as in your metabolic homeostasis – or the internal balance maintained by your body. Imbalances in thyroid levels lead to hypothyroidism or hyperthyroidism, showing just how important balance is in the system. Madrigal Pharmaceutical has been working on synthesizing TH analogs – which are compounds that have a similar molecular structure as TH and can therefore interact with at least some of its molecular targets.

Through synthesis of specific pyridazinone compounds, Madrigal Pharmaceuticals developed TH analogs and their associated prodrugs. The synthesis method developed is scalable while maintaining the purity, suggesting it is a possible wide-scale solution. This analog is capable of reducing or eliminating the resulting negative effects of hypo or hyperthyroidism, while maintaining the interactions typically needed by TH. The development of thyroid hormone analogs would open new avenues of treatment for patients with metabolic and thyroid related diseases.
