New Mexico Patent of the Month – March 2022

Antibodies are produced by specialized white blood cells. In an effort to speed up drug discovery pipelines, the scientific community has developed antibody libraries which provide a searchable database of existing antibodies. There are two main kinds of libraries – synthetic and natural antibody libraries. Specifica, Inc. is part of this endeavor, developing libraries to revolutionize antibody discovery.

Specifica’s libraries are made of a series of both heavy chain variable domains and light chain variable domains. These each have complementary determining regions (CDR) found in naturally-occurring human antibodies and the methods of making them. Their libraries are constructed in a way that ensures it holds a unique combination of naturally replicated CDRs as well as at least one hundred million unique HCDR3s (heavy chain complementarity determining region 3) from donor pools.  HCDR3s are amplified directly from purified B cells, while replicated natural CDRs, derived from Specifica’s databases, are used for the rest. The exclusive use of combinatorially reassembled natural CDR sequences, rather than intra-CDR synthetic diversity, ensures correct folding, avoids co-variance violations, and allows the complete elimination of sequence based liabilities for five of the six CDRs.

This means each library generated is completely unique. By increasing the diversity within the antibody libraries, the quality of antibodies are also increased. Many other libraries have been limited by this need for diversity.
